Biochemical taxonomy of Crenicichla (Pisces: Perciformes: Cichlidae) of the Iguaçu River, Brazil BABT

Samples of Crenicichla iguassuensis and a morph from the Iguaçu River which differed from C. iguassuensis by its lips morphology were examined by isozyme starch electrophoresis technique. The analysis of 27 enzyme loci exhibited a genetic identity of 0.993 between the two morphs. No diagnostic locus was found that permitted to distinguish the two Crenicichla morphs. The data obtained in this investigation did not support the existence of two distinct species for this genus in the Iguaçu River, which suggested that the morphs stood for two polymorphic forms of Crenicichla iguassuensis.

Microsatellite markers for identification of a group of italian olive accessions Scientia Agricola

Cultivar characterization for fruit trees certification requires fast, efficient and reliable techniques. Microsatellite markers (SSR) were used in the molecular characterization of 23 genotypes of Olea europaea subsp europaea. The DNA from the olive cultivars was analyzed using nine pre-selected SSR primers (GAPU59, GAPU71A, GAPU71B, GAPU103A, UDO99-01, UDO99-12, UDO99-28 and UDO99-39) and revealed 29 alleles, which allowed each genotype to be identified. In the dendrogram, the nine primers allowed the 23 olive genotypes to be grouped into subgroups corresponding to the same cultivar denominations. SSR markers proved to be efficient and reliable for the molecular characterization of Italian olive cultivars.

Molecular taxonomy of Plagioscion Heckel (Perciformes, Sciaenidae) and evidence from mtDNA RFLP markers for an invasive species in the Paraná river, Southern Brazil Zoologia

Mitochondrial RFLP markers were developed to examine whether Plagioscion squamosissimus (Heckel, 1840) is invasive in natural environments of the congener P. ternetzi in the Paraná river, in southern Brazil. Specimens of P. squamosissimus and of the putative P. ternetzi (Boulenger, 1895) were obtained from the Negro river (Manaus, Amazonas, Brazil) and from Paraná river, respectively. Fragments of the cytochrome b gene (900bp) were amplified by PCR and four restriction enzymes (Eco RI, Mbo I, Bam HI and Alu I) yielded the mitochondrial markers. An additional RFLP analysis with a cytochrome b gene sequence of Plagioncion sp. from GeneBank was carried out to validate the prior analysis.
